FIS-B contains the column of winds aloft directly above the tower. Since that's only useful for the immediate area to the tower, most EFBs require the full picture of winds aloft along the route to give useful planning numbers, which needs an internet connection. You can see the ADS-B provided winds in iFly by tapping near the tower to open the QuickMenu, switching to the WX tab if not selected, hitting the WX Info... button, and switching to Winds.

Yes, you can use the method below to update using your SD card, or a USB thumb drive, without unplugging anything.

As others have said, all you need to do is pull the SD card and update it from a PC/laptop if you want to leave the 740 in the plane.  You can also go old-school and create an update package on a USB thumb drive and again do your update in the plane, though that method will take longer.

However, round-barrel power connectors were designed for easy unplugging/plugging, and millions of devices have been using various iterations of such connectors for years, with jillions of cycles and low failure rates.  With normal use, there will be no problems.  SD card and USB ports are also designed for frequent usage and long cycle lives.  The antenna connectors are a little more delicate and not as common, so I don't know about those...but do you really need an external antenna?  My 720 gets great GPS reception just from its built-in receiver, though nowadays I feed it with a Stratux and so the internal GPS is not used.

There are lots of things to stay up nights worrying about.  Wearing out your 740b because you take it home after every flight is not one of them. 

I take mine home after every flight just because I prefer to have it with me instead of sitting in the plane.

 I'd never heard that the ADS-B supplied winds aloft were tied to the tower location.  I'd always thought the winds/temps aloft info was the same NOAA/FAA winds you get from official FAA sources, and reported for the same airports as the FAA sources.  Is iFly doing some interpolation to get the winds at the tower, or is it broadcast that way?  I haven't looked at the ADS-B winds aloft info (while flying) in quite some time.

That's because I misunderstood the explaination I got. I apologize. I did some more digging and found the correct answer. The FIS-B broadcast includes all weather stations within a 1000NM radius of the ADS-B tower. Those weather stations show the column directly above them, updated every 6 hours.
